The current model of diabetes and its complications strongly suggests that hyperglycemia is the main factor involved in the development of diabetic vascular disease. High glucose levels trigger a chain of events leading to increased endothelial permeability and an altered barrier function. The molecular mechanism(s) linking hyperglycemia to endothelial dysfunction remain(s) to be elucidated. Given the pivotal role of vascular endothelium in blood fluid homeostasis, we hypothesized that hyperglycemia may affect endothelial barrier function by altering the expression of tight junction proteins, which form the paracellular pathway. To address this hypothesis, we compared tight junction proteins expression and localization in human umbilical vein endothelial cells (HUVEC) grown in culture under normoglycemic and hyperglycemic conditions. We found that HUVEC grown in hyperglycemia display increased endothelial permeability and reduced expression of ZO-1, occludin and claudin-5 proteins, which is associated with a decrease in the total amount of ZO-1, occludin and claudin-5 protein. However, hyperglycemia did not significantly affect the expression of claudin-3, -4, or -8 proteins. To characterize further the cellular mechanisms responsible for altered expression of ZO-1, occludin and claudin-5 in hyperglycemia, we assessed the effects of glucose on the phosphorylation of tight junction proteins. Hyperglycemia enhanced the phosphorylation of ZO-1, occludin and claudin-5, suggesting that the altered protein expression in hyperglycemia may be attributed to a post-translational modification of these tight junction proteins.
